
三叶青的化学成分及其体外抗肿瘤活性研究
Chemical Constituents of Whole Plants of Tetrastigma hemsleyanum and Their Antitumor Activities
目的 研究三叶青全草的化学成分及其体外抗肿瘤活性。方法 运用多种色谱技术分离三叶青中化学成分,采用1H-NMR、13C-NMR、MS、HR-MS等波谱技术确定化合物结构。以噻唑蓝法,测试各黄酮化合物对HepG2,HCT-8,A549 3种肿瘤细胞的体外抑制作用。结果 分离鉴定13个化合物,分别为β-谷甾醇(1)、十六烷酸(2)、十七烷酸(3)、芹菜素(4)、槲皮苷(5)、山柰酚-3,7-O-L-二鼠李糖苷(6)、山柰酚-3-O-新橙皮糖苷(7)、芹菜素-6-C-β-D-吡喃葡萄糖苷(8)、芹菜素-8-C-α-L-吡喃鼠李糖-(1-2)-β-D-吡喃葡萄糖苷(9)、芹菜素-8-C-β-D-吡喃葡萄糖苷(10)、芹菜素-8-C-β-D-吡喃葡萄糖-(1-4)-β-D-吡喃葡萄糖苷(11)、荭草素(12)、异荭草素(13)。结论 化合物4~6,8~13系首次从该植物中分离得到,体外噻唑蓝活性筛选表明三叶青黄酮类化合物具有一定的抗肿瘤细胞增殖作用。
OBJECTIVE To study the chemical constituents and antitumor activity of Tetrastigma hemsleyanum Diels et Gilg. METHODS The compounds were isolated by various column chromatography. Their structures were identified by spectroscopic methods including 1H-NMR, 13C-NMR, MS, HR-MS, etc. The antiproliferative activity of the purified flavonoids compounds were evaluated by MTT assay against HepG2, HCT-8, and A549 tumor cells. RESULTS Thirteen compounds were isolated and identified as β-sitosterol (1), palmitic acid (2), heptadecanoic acid (3), apigenin (4), quercitrin (5), kaempferitrin (6), kaempferol-3-O-neohesperidoside (7), apigenin-6-C-β-D-glucopyranoside (8), apigenin-8-C-α-L-rhamnopyranosyl-(1-2)-β-D-glucopyranoside (9), apigenin-8-C-β-D-glucopyranoside (10), apigenin-8-C-β-D-glucopyranoside-(1-4)-β-D-glucopyranoside (11), orientin (12), and isoorientin (13). CONCLUSION Compounds 4-6 and 8-13 were isolated from T. hemsleyanum for the first time. The flavonoids of T. hemsleyanum showed inhibitory activity on HepG2, HCT-8, and A549 tumor cells.
